Abstract: |
The processes of scallop meats can produce a large amount of by-products like scallop skirts, which are rich in nutrients. This work aimed to obtain the information about antioxidant peptides derived from scallop skirts of Mimachlamys nobilis. The hydro-lysates obtained by trypsin exhibited the highest antioxidant activity (75.6%, 79.5% and 76.7% using DPPH, hydroxyl and superoxide anion radical scavenging activity assays, respectively). The antioxidant peptides (MNAP) were separated and purified using UF membranes, GFC processes and RP-HPLC, and the antioxidant activity of MNAP was identified to be 85.7% using DPPH radical scavenging activity assay. The peptides (MNAP) were composed of two peptides with their molecular weights 2 855.6 Da and 3 007.2 Da using MALDI-TOF-MS, respectively. The results suggested that the scallop skirts of Mimachlamys nobilis could be explored as a source of antioxidant peptides. |

华贵栉孔扇贝裙边抗氧化活性肽的纯化与鉴定 王全富1,王一帆1,侯艳华1 ,徐一凤1,闫美宏1,李昊远1,刘远平2,李钰金3 (1哈尔滨工业大学(威海),海洋科学与技术学院,山东 威海 264209, 2山东省海洋健康食品工程技术研究中心,山东 荣成 264300 3山东海洋功能食品发展联合实验室,山东 荣成 264300) 创新点说明:1)本文利用含有丰富营养物质的加工副产物扇贝裙边进行了抗氧化活性肽的纯化相关研究。 2)本文利用多级纯化的方法与多种抗氧化活性评价方法测定了扇贝裙边抗氧化活性肽的分离与鉴定,最终获得了具有极高抗氧化能力的小分子生物活性肽。 3)本研究对于海洋生物活性肽的工业制备与实际生产开发与应用提供了重要的理论依据。 研究目的: 以华贵栉孔扇贝裙边为原料分离纯化得到抗氧化活性较高的生物活性肽。 研究方法: 利用5种蛋白酶酶解华贵栉孔扇贝裙边,筛选得到酶解液抗氧化能力最强的蛋白酶。以DPPH自由基清除率、羟基自由基清除率与超氧阴离子自由基清除率为指标,利用超滤、凝胶柱层析、SDS-PAGE与反相高效液相色谱对扇贝裙边酶解液进行分离纯化,以获得高纯度抗氧化活性肽。利用质谱法鉴定最终纯化得到的抗氧化活性肽的分子量。 结果: 1)胰蛋白酶解扇贝裙边得到的酶解液的抗氧化活性最高,其DPPH自由基清除率、羟基自由基清除率与超氧阴离子自由基清除率分别为75.6 %,79.5 %与76.7 %。 2)利用多级纯化得到的抗氧化活性肽命名为MNAP,其DPPH自由基清除率为85.7 %,质谱法测定MNAP的包含2个小肽,其分子量分别为2 855.6 Da与3 007.2 Da。 结论: 华贵栉孔扇贝裙边能够作为用于制备具有较强抗氧化能力的生物活性肽。 关键词:华贵栉孔扇贝;抗氧化活性肽;纯化;扇贝裙边;水解产物 |
