摘要: |
为明确不同提取方法获得的玉米麸皮多糖对人结肠癌HT-29细胞增殖的抑制作用,采用MTT、倒置显微镜观察、分裂指数、集落形成方法测定细胞活力.MTT实验结果表明,水提玉米麸皮多糖可抑制结肠癌HT-29细胞的生长,随质量浓度和处理时间的增加细胞抑制率增加,0.02 g/L的多糖处理48 h其抑制率可达60.62%.另外两种多糖对HT-29细胞无明显的抑制效果.形态学观察结果表明,水提玉米麸皮多糖作用48 h,HT-29细胞有明显的形态学改变.同时,水提玉米麸皮多糖能够抑制HT-29细胞有丝分裂数和集落形成率,并呈时间和剂量效应. |

Abstract: |
To investigate the inhibitory effect of polysaccharides from corn bran extracted by three different methods on the HT-29 cells, it was studied by MTT assay, inverted microscope, mitotic index and colony formation. MTT results showed that the water extraction polysaccharide from corn bran had a significant inhibition effect on the proliferation of HT-29 cells in a dose-dependent manner. After treatment for 48 h with 0.02 g/L of water extraction polysaccharide from corn bran, the inhibitory rate was 60.62%. The other two polysaccharides had no affect on the growth of HT-20 cells. The morphologic observation showed that the water extraction polysaccharide from corn bran induced obvious morphological changes. The mitotic index and colony formation was decreased compared with the control group as the incubation time and concentration of water extraction polysaccharide from corn bran. |
